FITC is most commonly-used fluorescent dye for FACS analysis. FITC is a small organic molecule, and is typically conjugated to proteins via primary amines (i.e., lysines). Usually, between 3 and 6 FITC molecules are conjugated to each antibody; higher conjugations can result in solubility problems as well as internal quenching (and reduced brightness). Thus, an antibody will usually be conjugated in several parallel reactions to different amounts of FITC, and the resulting reagents will be compared for brightness (and background stickiness) to choose the optimal conjugation ratio. Fluorescein is typically excited by the 488 nm, and emission is collected at 530 nm.

Biuret is a small compound that forms when urea is heated allowing two urea molecules to join. Urea molecules fused in this manner produce amide groups (-NH) at the center of the molecule which bind to cupric ions at a basic pH. The copper complexes that result from this interaction produce a strong blue color that can be measured with a spectrophotometer. Proteins also contain amide groups. When an amino group and a carboxyl group join to form a peptide bond, the amino group (-NH2) becomes an amide group (-NH). At basic pH, the proteins forms complex with copper ions. Because this reaction was first observed with biuret, it is called the biuret reaction, and when this reaction is used to measure protein concentrations, it is called the Biuret Protein Assay.

Western blotting is an analytical method wherein a protein sample is electrophoresed on an SDS-PAGE and electrotransferred onto nitrocellulose membrane. Chicken antibodies isolated from egg yolks (IgY) are a suitable alternative to conventional antibodies from blood of experimental animals. Main advantages of IgY are: (i) larger amounts of IgY could be prepared from repeatedly laid chicken eggs, (ii) collection of eggs is a non-invasive procedure contrary to bleeding of mammals, and (iii) a better response of chicken to mammalian antigens due to the larger evolutionary distance. In our procedure easy, cost- and labor-effective two-step procedure are followed, which consisting of yolk extraction by tap water followed by a specific precipitation of IgY at pH 4.0 with sodium chloride at the final concentration of 8.8%. The final protein product obtained was analyzed with SDS-PAGE.

Alkaline phosphatase (ALP) catalyzes the hydrolysis of phosphate esters in alkaline buffer and produces an organic radical and inorganic phosphate. Changes in alkaline phosphatase level and activity are associated with various disease states in the liver and bone. Kit uses p-nitrophenyl phosphate (pNPP) as a phosphatase substrate which turns yellow (λmax= 405 nm) when dephosphorylated by ALP. The Kit can detect 10-250 µU ALP in samples. Bacterial Alkaline Phosphatase (BAP) is a phosphomonoesterase that hydrolyzes 3′ and 5′ phosphates from DNA and RNA. It is suitable for removing 5′ phosphates prior to end-labeling and for dephosphorylating vectors prior to insert ligation. BAP is active at 65 DegreeC for at least 1 h. It is sensitive to inhibition by micromolar amounts of inorganic phosphate. The enzyme is purified from E. coli antibody HRP conjugate .Horseradish Peroxidase is coupled to IgG antibody in a two-step procedure. In the first step monosaccharide residues in the enzyme are oxidized with periodate to produce aldehyde groups. Then, in the second step, the aldehyde groups are allowed to react with amino groups in the IgG antibody. The Schiff bases formed are reduced and the conjugate is purified by dialysis.

Enzyme kinetics is the study of the chemical reactions that are catalysed by enzymes. In enzyme kinetics, the reaction rate is measured and the effects of varying conditions of the reaction are investigated. Studying an enzyme's kinetics in this way can reveal the catalytic mechanism of this enzyme, its role in metabolism, how its activity is controlled, and how a drug or an agonist might inhibit the enzyme. Enzymes are usually protein molecules that manipulate other molecules the enzymes' substrates. These target molecules bind to an enzyme's active site and are transformed into products through a series of steps known as the enzymatic mechanism. These mechanisms can be divided into single-substrate and multiplesubstrate mechanisms. Kinetic studies on enzymes that only bind one substrate, such as triosephosphate isomerase, aim to measure the affinity with which the enzyme binds this substrate and the turnover rate. Some other examples of enzymes are phosphofructokinase and hexokinase, both of which are important for cellular respiration (glycolysis). SDS-PAGE, sodium dodecyl sulfate polyacrylamide gel electrophoresis, is a technique used in biochemistry, genetics and molecular biology to separate proteins according to their molecular weight. The electrophoretic mobility of proteins depends upon their size. As proteins are amphoteric compounds, their net charge can therefore be determined by the pH of the medium in which they are suspended.
