The serum protein electrophoresis (SPEP) test measures specific proteins in the blood to help identify some diseases. Proteins are substances made up of smaller building blocks called amino acids. Proteins carry a positive or a negative electrical charge, and they move in fluid when placed in an electrical field. Serum protein electrophoresis uses an electrical field to separate the proteins in the blood serum into groups of similar size, shape, and charge. Blood serum contains two major protein groups: albumin and globulin. Both albumin and globulin carry substances through the bloodstream. Using protein electrophoresis, these two groups can be separated into five smaller groups (fractions).

SDS-PAGE, sodium dodecyl sulfate polyacrylamide gel electrophoresis, is a technique used in biochemistry, genetics and molecular biology to separate proteins according to their molecular weight. The electrophoretic mobility of proteins depends upon their size. As proteins are amphoteric compounds, their net charge can therefore be determined by the pH of the medium in which they are suspended. Therefore, at a given pH and under non-denaturing conditions, the electrophoretic separation of proteins is determined by both size and charge of molecules. As proteins are high molecular weight molecules, it needs porous gels to get separated. Polyacrylamide gels are those which provide a means of separating proteins by size as they are porous.

Western blotting is an analytical method wherein a protein sample is electrophoresed on an SDS-PAGE and electrotransferred onto nitrocellulose membrane. The transferred protein is detected using specific primary antibody and secondary enzyme labeled antibody and substrate. A protein sample is subjected to polyacrylamide gel electrophoresis. After this the gel is placed over a sheet of nitrocellulose and the protein in the gel is electrophoretically transferred to the nitrocellulose. The nitrocellulose is then incubated with the specific antibody for the protein of interest. The nitrocellulose is then incubated with a second antibody, which is specific for the first antibody.

Alkaline phosphatase (ALP) catalyzes the hydrolysis of phosphate esters in alkaline buffer and produces an organic radical and inorganic phosphate. Changes in alkaline phosphatase level and activity are associated with various disease states in the liver and bone. Kit uses p-nitrophenyl phosphate (pNPP) as a phosphatase substrate which turns yellow (λmax= 405 nm) when dephosphorylated by ALP. The Kit can detect 10-250 µU ALP in samples. Bacterial Alkaline Phosphatase (BAP) is a phosphomonoesterase that hydrolyzes 3′ and 5′ phosphates from DNA and RNA. It is suitable for removing 5′ phosphates prior to end-labeling and for dephosphorylating vectors prior to insert ligation. BAP is active at 65 DegreeC for at least 1 h. It is sensitive to inhibition by micromolar amounts of inorganic phosphate. The enzyme is purified from E. coli antibody HRP conjugate .Horseradish Peroxidase is coupled to IgG antibody in a two-step procedure. In the first step monosaccharide residues in the enzyme are oxidized with periodate to produce aldehyde groups. Then, in the second step, the aldehyde groups are allowed to react with amino groups in the IgG antibody. The Schiff bases formed are reduced and the conjugate is purified by dialysis.

FITC is most commonly-used fluorescent dye for FACS analysis. FITC is a small organic molecule, and is typically conjugated to proteins via primary amines (i.e., lysines). Ion exchange chromatography separates molecules based on differences between the overall charges of the proteins. This method utilizes a combination of DNase and DNA Polymerase to nick one strand of the DNA helix, then incorporate labeled nucleotides as the polymerase examines, or “proofreads” the nicked site. Native PAGE uses the same discontinuous chloride and glycine ion fronts as SDS-PAGE to form moving boundaries that stack and then separate polypeptides by charge to mass ratio. Therefore you can easily see multiple bands from the camshot of your native PAGE gel if your target protein has polymerized forms in your sample. In native PAGE electrophoresis most proteins have an acidic or slightly basic pl(isoelectric point) (~3–8) and migrate towards the negative polar.

Chicken antibodies isolated from egg yolks (IgY) are a suitable alternative to conventional antibodies from blood of experimental animals. Main advantages of IgY are: (i) larger amounts of IgY could be prepared from repeatedly laid chicken eggs, (ii) collection of eggs is a non-invasive procedure contrary to bleeding of mammals, and (iii) a better response of chicken to mammalian antigens due to the larger evolutionary distance. In our procedure easy, cost- and labor-effective two-step procedure are followed, which consisting of yolk extraction by tap water followed by a specific precipitation of IgY at pH 4.0 with sodium chloride at the final concentration of 8.8%. The final protein product obtained was analyzed with SDS-PAGE.

Enzyme kinetics is the study of the chemical reactions that are catalysed by enzymes. In enzyme kinetics, the reaction rate is measured and the effects of varying conditions of the reaction are investigated. Biuret is a small compound that forms when urea is heated allowing two urea molecules to join. Urea molecules fused in this manner produce amide groups (-NH) at the center of the molecule which bind to cupric ions at a basic pH. The copper complexes that result from this interaction produce a strong blue color that can be measured with a spectrophotometer. Proteins also contain amide groups. When an amino group and a carboxyl group join to form a peptide bond, the amino group (-NH2) becomes an amide group (-NH). At basic pH, the proteins forms complex with copper ions. Because this reaction was first observed with biuret, it is called the biuret reaction, and when this reaction is used to measure protein concentrations, it is called the Biuret Protein Assay.

Silver staining is used to detect proteins after electrophoretic separation on polyacrylamide gels. It combines excellent sensitivity (in the low nanogram range) . The rationale of silver staining is quite simple. Proteins bind silver ions, which can be reduced under appropriate conditions to build up a visible image made of finely divided silver metal.
