DNA in the sample is released from blood cells using solution-I (Detergent). The cell wall of the blood cell is weakened by EDTA in the Solution-II. Solution-III denatures the cellular proteins. Extraction with phenol denatures the protein, Chloroform removes SDS- Protein / polysaccharide complexes as white interface between the aqueous and organic phase after centrifugation and isoamyl alcohol prevents foaming. The high molecular weight genomic DNA in aqueous phase is precipitated with 100 % ethanol or isopropanol and further the salts are removed by washing with 70% alcohol.

In molecular biology, restriction fragment length polymorphism, or RFLP (commonly pronounced “rif-lip”), is a technique that exploits variations in homologous DNA sequences. It refers to a difference between samples of homologous DNA molecules that come from differing locations of restriction enzyme sites, and to a related laboratory technique by which these segments can be illustrated. A very simple, fast, universally reproducible method to extract high quality megabase genomic DNA from different organisms is described. This same method is applicable to extract high quality complex genomic DNA. The method does not require expensive and environmentally hazardous reagents and equipment. DNA ligation is the process of joining together two DNA molecules ends (either from the same or different molecules). Specifically, it involves creating a phosphodiester bond between the 3' hydroxyl of one nucleotide and the 5' phosphate of another. This reaction is usually catalyzed by a DNA ligase enzyme. This enzyme will ligate DNA fragments having blunt or overhanging, complementary, 'sticky' ends. Typically, it is easier to ligate molecules with complementary sticky ends than blunt ends. T4 DNA ligase is the most commonly used DNA ligase for molecular biology techniques and can ligate 'sticky' or blunt ends. After ligation, the insert DNA is physically attached to the backbone and the complete plasmid can be transformed into bacterial cells for propagation. Most restriction enzymes digest DNA asymmetrically across their recognition sequence, which results in a single stranded overhang on the digested end of the DNA fragment. The overhangs, called "sticky ends", are what allow the vector and insert to bind to each other. When the sticky ends are compatible, meaning that the overhanging base pairs on the vector and insert are complementary, the two pieces of DNA connect and ultimately are fused by the ligation reaction.

Plasmid is a double stranded, circular extra chromosomal DNA of bacterium. It is used in recombinant DNA experiments to clone genes from other organisms and make large quantities of their DNA. Plasmid can be transferred between same species or between different species. Size of plasmids range from 1-1000 kilo base pairs. Nested PCR used to increase the specificity of DNA amplification. Two sets of primers are used in two successive reactions. In the first PCR, one pair of primers is used to generate DNA products, which may contain products amplified from non-target areas. The products from the first PCR are then used as template in a second PCR, using one ('hemi-nesting') or two different primers whose binding sites are located (nested) within the first set, thus increasing specificity. Nested PCR is often more successful in specifically amplifying long DNA products than conventional PCR, but it requires more detailed knowledge of the Sequence of the target.

The vectors used in cloning are manipulated in such a way that this α-complementation process serves as a marker for recombination. A multiple cloning site (MCS) is present within the lacZ sequence in the plasmid vector. This sequence can be nicked by restriction enzymes to insert the foreign DNA. When a plasmid vector containing foreign DNA is taken up by the host E. coli, the α-complementation does not occur, therefore, a functional β-galactosidase enzyme is not produced. If the foreign DNA is not inserted into the vector or if it is inserted at a location other than MCS, the lacZ gene in the plasmid vector complements the lacZ deletion mutation in the host E. coli producing a functional enzyme.

This is done using an agarose gel electrophoresis of known molecular weight nucleic acids (ladder) along with the protein or nucleic acid to be characterized. A linear relationship exists between the logarithm of the molecular weight of native nucleic acid, and it’s Rf. The Rf is calculated as the ratio of the distance migrated by the molecule to that migrated by a marker dye-front.
A simple way of determining relative molecular weight by electrophoresis is to plot a standard curve of Rf vs. log Mw for known samples (ladder), and read off the log Mw of the sample after measuring distance migrated on the same gel. After introducing the data of ladder and sample Rf, sample Mw is calculated.
