    Descriptions:

    Mutations often inactivate a biosynthetic pathway of the microorganism, and frequently make a microorganism unable to grow on a medium lacking an adequate supply of the pathway’s end product. Based on this principle microorganism are classified as Prototrophic and Auxotrophic. Prototrophic organisms (wild type) have the same nutritional requirements as that of their ancestors. They need only inorganic salts, an organic energy source such as sugar, fat, protein and water to survive and grow. That is, the Prototroph need only "Minimal medium" for their growth and survival. Auxotrophic mutants are unable to grow without one or more essential nutrients. Auxotroph’s are mutant for particular nutrient synthesis pathway enzymes.Such an error is known as an inborn error of metabolism, whether it occurs in a bacterium or a eukaryote. An auxotroph can be grown only on an enriched medium that provides the particular nutrient that the mutant cannot metabolize on its own.

    Genomic DNA Extraction From Fungi Teaching Kit.

    We have developed a new, simple and effective method for extraction of fungal genomic DNA. The initial steps involved suspension of freeze-dried mycelium in buffer containing sodium dodecyl sulphate, detachment of DNA from polysaccharides by mild shearing, NaCl precipitation of polysaccharides and protein, chloroform extraction and ethanol precipitation.
    The ethanol precipitate was then subjected to a second round of mild shearing, NaCl precipitation, chloroform extraction and ethanol precipitation.
    The procedure required approximately 1 h to perform. The method yielded 8-32 microg of high molecular weight DNA per 30 mg of freeze-dried mycelium when tested on six fungal species.

    Methods for identifying isolates of various pathogenic bacteria by DNA fingerprinting with random primers (RAPD) have been described recently. In these methods many primers are screened and the primers that generate the most informative DNA pattern are selected. A new strategy that simplifies the primer selection process for RAPD fingerprinting has been developed in our laboratory. In this approach, one or more degenerate nucleotides is introduced into the core RAPD primer sequence at various nucleotide positions. Results show that a single degenerate nucleotide in the primer sequence can significantly change the DNA profile obtained for the same template.

      A procedure for the rapid isolation of DNA from the yeast Saccharomyces cerevisiae is described. To release plasmid DNA for the transformation of Escherichia coli , cells are subjected to vortex mixing in the presence of acid-washed glass beads, Triton X-100, sodium dodecyl sulfate, phenol and chloroform.
      Centrifugation of this mixture separates the DNA from cellular debris. E. coli can be efficiently transformed with plasmid present in the aqueous layer without further purification of the plasmid DNA.
      This procedure also releases chromosome DNA. Follo~ng two ethanol precipitations, the chromosomal DNA can be digested by restriction endonucleases poly A adenylation to mRNA.
